A cellular defence against lipid damage

GPX4 helps protect cells from damaging lipid oxidation. When that protection is insufficient, cells can undergo a form of death called ferroptosis. Researchers are investigating whether some cancer cells depend on this defence in ways that could be therapeutically useful. [M07]

What published work has shown

Published studies have reported GPX4-related sensitivity in particular glioblastoma cell states. These findings provide a reason to ask more focused questions; they do not establish a treatment for people with glioblastoma.

Our selected question

NEUVIOR proposes to examine whether a useful GPX4 dependency remains in malignant cells after senescent-cell depletion. The study needs to distinguish an additional survivor population from a stronger effect on the same population.

It must also consider normal neural systems. GPX4 has important roles in healthy cells, so the relevant question includes the separation between a tumour effect and unwanted effects elsewhere. [M08]

An intervention rationale would need converging evidence about mechanism, regrowth, exposure, and selectivity before further development.
